10.Summary of Research Results
|
The TERMINAL FLOWER 1 (TFL1) gene of Arabidopsis serves a key function in the development of the shoot apical meristem (SAM). The specific expression of TFL1 transcripts in the subapical region of the SAM suggests that TFL1 functions non-cell-autonomously and that TFL1 function may involve cell-cell communication. We demonstrate that the TFL1 protein is transmitted from the subapical region to the epidermis, and suggest that this protein trafficking is regulated during the developmental phase of the SAM. Our results suggest that TFL1 protein trafficking gives rise to the interlayer signaling responsible for the coordination of the distinct layers of the meristem. The L1-layer specific genes, PDF1 and PDF2 were isolated and studies. The PDF1 has the L1-box cis regulator. The PDF1 gene has the HD-GL2 class Homeo domain. This is suggested to be regulator of PDF1 gene by gel-shift assay. The transgenic plants carrying over-expression and co-suppressed PDF2 gene were isolated and studies. They showed various flowering phenotypes, which suggested the roles of L1-layer for flowering. Molecular genetic analysis of Arabidopsis FT locus has elucidated its role in integrating three genetic pathways regulating the floral transition. CONSTANS mediates signals from the photoperiod pathway by directly activating FT transcription. The autonomous and vernalization pathways act through FLOWERING LOCUS C which encodes a putative negative regulator of FT expression. FT acts in parallel with another pathway integrator, LEAFY, and in an antagonistic manner with its homolog, TERMINAL FLOWER1. We investigated functions of individual members of a gene family which encode endoxyloglucan transferases. What we found is as follows: 1) This multi-gene family consists of 33 genes in Arabidopsis. 2) individual genes exhibit spatially and physiologically distinct expression profiles, some members being involved in construction of cell plate and others in the primary and secondary cell walls. 3) EXGT-A3 gene disruptant showed morphological changes in the shoot of Arabidopsis. |
