１０．研究成果の概要
|
ニューロンにおけるシグナルトランスダクションのどの部分の異常がどのような行動異常に関係しているかについて、様々なノックアウトマウスを用いることにより明らかにした。用いたノックアウトマウスはNR2A(-/-)、NR2C(-/-)、NR2AC(-/-)、PLCβ4(-/-)、GFAP(-/-)であり、研究対象は小脳、海馬、視床内のニューロンであり、アッセイに用いた方法論としては連合学習（瞬目反射条件付け、恐怖学習）、概日リズム、睡眠、虚血性細胞死であった。本研究全体を記述するキーワードとしてはCaシグナリング機構が、またシグナリングを制御するキー分子としてPLCβ4と電位依存性Caチャネルに集約される。この結果Caシグナリングがニューロン機能の制御すなわち、LTD形成、リン酸化、トランスミッタ遊離、イオンチャネルコンダクタンス制御にどのように関わっているかがより一層明らかとなった。しかしながらPLCノックアウトマウスのニューロンの示すシグナルの変化は、これまで薬理学的にインヒビターを用いて行なわれた電気生理的、生化学的データからは説明できない場合が多々見受けられた。特にGタンパクからPLCへと伝達されるシグナルの行き先が失われたとき、Gタンパクによって直接開閉を制御されているイオンチャネルの開閉機構にも変化が見られた。これらはノックアウトマウスを用いて行なう研究に於いて注意を払うべきポイントとなろう。 一方また未来の研究発展のために以下の2つの方法論を開発した。即ち熱イメージングとCa感受性蛍光タンパクの細胞内での強制発現法の開発である。この結果細胞内シグナリングを熱力学的な見地から扱えるようになった。また細胞内の任意の小器官や特定のチャネルの近傍のCa濃度が測定できるようになった。 |
