10.Summary of Research Results

Our research objective is the regeneration of damaged tissues and organs using a new in situ tissue engineering approach. This method aims to achieve autologous tissue and organ regeneration, and is comprised of the steps of:
・ implantation of a 3-dimentional scaffold made of reconstituted collagen and bioabsorbable synthetic polymer,
・ addition of a DDS consisting of cell growth factors, and
・ when necessary, seeding autologous precursor cells or interesitial stem cells taken from autologous bone marrow.
Results: (1) membranous tissues (eg. dura matter), (2) blood vessels, (3) the trachea, (4) the esophagus, (5) permanent teeth and periodontal tissue, (6) bone, (7) and segments of peripheral nerves upto 8cm in length have been successfully regenerated without the need for cell seeding. Successful regeneration has also been achieved in (8) the stomach, intestine, heart, and bladder, but cell seeding was required for full regeneration of proper muscle components. Partial connection of the (9) spinal cord has been achieved without cell seeding, and thus experimentation is continuing to investigate autologous neuron implantation. In (10) the lung, regenerated small bronchi were observed to appear. In (11) the liver, a regenerated, healthy liver structure with new ductal tubes and blood vessels was observed.
