10.Summary of Research Results
|
This project if constructed from 3 main research fields, i.e., 1) reconstruction of hepatic functions, 2) reconstruction of bone and joint functions, and 3) reconstruction of neo-vascular functions. Results obtained in each fields are described below. In the research for the reconstruction of hepatic functions, development of a novel bioartificial liver (BAL), packed-bed type BAL, was attempted. This series of research included scaling up of the BAL, optimization of culture conditions, establishment of a hepatic failure model pig, determination of the conditions of extracorporeal circuit, and application of fetal liver cells as a cell source of the BAL. Moreover, to the purpose of the establishment of new treatment of liver failure by cell transplantation or of clarifying the mechanism of liver regeneration, identification of hepatic stem cells were performed by use of FACS analysis. In the reconstruction of bone and joint functions, bone formation by osteoblast and chondro-induction from bone marrow cells were investigated. To reconstruct the hematopoietic functions in bone marrow, ex vivo expansion and proliferation of hematopoietic stem cells as well as basic studies clarifying mechanisms of differentiation and proliferation of hematopietic cells were performed. In the reconstruction of neo-vascular functions, mechanism of recovery of endothelial cells after vascular injury were investigated. Moreover, a new PTCA device which can prevent vascular damage after PTCA treatment, was developed. Establishment of transgenic rabbits which are effective models to clarify the mechanism of atherogenesis, investigations of vascularization during tumor growth and of functions in blood-brain barrier, were also attempted. |
